Amlokos 5mg Tablet 10s is an oral medication belonging to the class of dihydropyridine Calcium Channel Blockers (CCBs). It is widely used to treat high blood pressure (hypertension) and certain types of chest pain (angina). It works by relaxing the blood vessels, allowing blood to flow more easily, which lowers blood pressure and reduces the workload on the heart. The standard starting and maintenance dose is typically 5mg, taken once daily.

Amlokos 5mg Tablet 10s (amlodipine) is a long-acting dihydropyridine calcium channel blocker used to treat high blood pressure (hypertension) and angina (chest pain). It works by blocking calcium entry into the smooth muscle cells of blood vessels, which causes the vessels to relax and widen. This lowers blood pressure and reduces the workload on the heart.
In hypertension, amlodipine primarily acts on peripheral arteries, decreasing vascular resistance and improving blood flow. Its gradual onset and long duration of action provide stable, 24-hour blood pressure control with once-daily dosing, helping to reduce day-to-day blood pressure fluctuations.
In angina, amlodipine improves blood supply to the heart by dilating the coronary arteries and preventing coronary artery spasm. This increases oxygen delivery to the heart muscle and helps reduce the frequency and severity of chest pain. It is effective in both chronic stable angina and vasospastic (Prinzmetal’s) angina.
Amlodipine may be used alone or in combination with other cardiovascular medicines, such as ACE inhibitors, beta blockers, or diuretics. It is generally well tolerated, with common side effects related to vasodilation, such as ankle swelling, flushing, or headache. Because of its proven effectiveness, safety profile, and convenient once-daily dosing, Amlokos 5mg Tablet 10s is widely used for long-term cardiovascular management.
To lower blood pressure and reduce the risk of fatal and nonfatal cardiovascular events, primarily strokes and myocardial infarctions.
o relieve symptoms of chest pain associated with physical exertion.
To prevent episodic, recurrent chest pain caused by temporary spasms of the coronary arteries.
Provides consistent, prolonged blood pressure control due to its long half-life.
Reduces the workload on the heart and improves oxygen supply to the cardiac muscle, beneficial for angina patients.
Simplifies treatment regimens and improves patient adherence.
Effective across various patient populations, including the elderly and those with diabetes or chronic kidney disease.
Amlokos 5mg Tablet 10s is generally well-tolerated, but some side effects may occur.
Amlokos 5mg Tablet 10s is a peripheral arterial vasodilator that acts directly on vascular smooth muscle.
Calcium Channel Blockade: It selectively blocks the entry of calcium ions (Ca2+) into vascular smooth muscle and cardiac muscle cells. Specifically, it acts on the L-type calcium channels.
Vasodilation: By limiting the influx of calcium, Amlokos 5mg Tablet 10s inhibits the contraction of arterial smooth muscles. This causes the arteries throughout the body (peripheral arteries) to relax and widen (vasodilation).
Blood Pressure Reduction: The widening of arteries reduces the total peripheral vascular resistance (the resistance the heart pumps against). This reduction effectively lowers systemic blood pressure.
Angina Relief: In the heart, it causes coronary vasodilation (widening of the arteries supplying the heart), increasing oxygen delivery. It also reduces peripheral resistance, decreasing the heart's workload and oxygen demand, thereby preventing and treating angina.
The typical starting daily dose is 5mg taken once daily. The dose can be adjusted by a healthcare professional up to a maximum of 10mg once daily.
In the event of an overdose, the main effect is pronounced peripheral vasodilation leading to severe and prolonged systemic hypotension and possibly reflex tachycardia (rapid heart rate). Action Required: Seek immediate emergency medical attention. Treatment is supportive, focusing on cardiovascular maintenance, including active monitoring, possible gastric lavage, and use of intravenous fluids and vasopressors if necessary.
If you miss a dose, take it as soon as you remember unless it is almost time for your next dose (more than 12 hours late). If it is close to the next dose, skip the missed one and continue your regular schedule. Do not take two doses at once.
