Olnyd Trio 40 Tablet contains chlorthalidone, cilnidipine, and olmesartan medoxomil, a triple combination used in the management of hypertension. Chlorthalidone is a thiazide-like diuretic that reduces fluid volume, cilnidipine is a calcium channel blocker that relaxes vascular smooth muscle, and olmesartan medoxomil is an angiotensin II receptor blocker that prevents vasoconstriction. Together they provide complementary blood pressure lowering through three distinct mechanisms. Always use Olnyd Trio 40 Tablet exactly as directed by your doctor.
Olnyd Trio 40 Tablet is a fixed-dose triple antihypertensive combination targeting blood pressure through diuretic, calcium channel blocking, and renin-angiotensin system inhibitory mechanisms.
Chlorthalidone is a long-acting thiazide-like diuretic that inhibits sodium and chloride reabsorption in the distal convoluted tubule of the kidney, increasing urinary sodium and water excretion. This reduces plasma volume and cardiac preload, lowering blood pressure.
Cilnidipine is a dual L-type and N-type calcium channel blocker. L-type blockade relaxes arterial vascular smooth muscle, reducing peripheral vascular resistance. N-type blockade additionally suppresses sympathetic noradrenaline release at nerve terminals, providing a more balanced antihypertensive effect with less reflex tachycardia compared to other dihydropyridine calcium channel blockers.
Olmesartan medoxomil is a prodrug that is converted to olmesartan after absorption. It selectively blocks AT1 angiotensin II receptors, preventing the vasoconstriction, aldosterone secretion, and sodium retention triggered by angiotensin II. This provides sustained blood pressure reduction and end-organ protection.
Together these three mechanisms provide additive and complementary antihypertensive effects, allowing effective blood pressure control with lower doses of each individual component.
The uses of Olnyd Trio 40 Tablet are as follows:
Olnyd Trio 40 Tablet is used to manage moderate to severe hypertension where monotherapy or dual therapy has not achieved adequate blood pressure control.
Chlorthalidone addresses the fluid retention component of hypertension in susceptible patients.
Cilnidipine's N-type calcium channel blockade provides additional benefit in patients with high sympathetic tone.
Olmesartan provides renal protection alongside blood pressure control in patients with diabetic kidney disease.
The triple combination is particularly useful in patients whose blood pressure remains uncontrolled on dual therapy.
Here are the benefits of Olnyd Trio 40 Tablet:
Like all medicines, Olnyd Trio 40 Tablet may cause side effects in some individuals.
Always consult your doctor if side effects persist or worsen.
To ensure safe use:
Olnyd Trio 40 Tablet lowers blood pressure through three complementary mechanisms.
Chlorthalidone inhibits the sodium-chloride cotransporter in the distal convoluted tubule, increasing urinary excretion of sodium and water. This reduces circulating blood volume and cardiac output, lowering blood pressure.
Cilnidipine blocks both L-type calcium channels in arterial smooth muscle, causing vasodilation and reduced peripheral resistance, and N-type calcium channels at sympathetic nerve terminals, suppressing noradrenaline release and reducing sympathetic vascular tone.
Olmesartan selectively and competitively blocks AT1 receptors, preventing angiotensin II from causing vasoconstriction, aldosterone release, and sodium retention. This produces sustained vasodilation and reduces both preload and afterload on the heart.
Inform your doctor of any known allergy to thiazide diuretics, calcium channel blockers, or angiotensin receptor blockers before use.
This combination is not recommended during breastfeeding. Consult your doctor for a suitable alternative.
Olmesartan is contraindicated in pregnancy due to risk of foetal harm. Discontinue immediately if pregnancy is detected.
Dizziness and hypotension, particularly during early treatment, may impair driving ability.
Alcohol may enhance the blood pressure lowering effect and worsen dizziness and hypotension.
Use carefully in hepatic impairment as metabolism of olmesartan and cilnidipine may be affected. Consult your doctor.
Renal function and electrolytes should be monitored regularly. Use carefully in significant renal impairment.
Safety and dosing in children has not been fully established. Use only if advised by a specialist.
Elderly patients are at increased risk of hypotension, electrolyte disturbance, and renal impairment. Close monitoring is advised.
A few practical measures can help improve treatment outcomes and ensure safe use of Olnyd Trio 40 Tablet:
Proper storage is important to maintain the stability and effectiveness of Olnyd Trio 40 Tablet:
Olnyd Trio 40 Tablet may generally be taken without strict dietary restrictions. However:
Olnyd Trio 40 Tablet may interact with the following medicines:
Olnyd Trio 40 Tablet should be used carefully in the following conditions:
Usually taken as a single oral dose once daily as directed by your doctor. Dose depends on the specific formulation strength and the degree of blood pressure control required.
If a dose is missed, take it as soon as remembered. Do not double the dose to make up for a missed one.
