How Estradiol and Dydrogesterone Work Estradiol and Dydrogesterone is a combination of two hormonal medications used in hormone replacement therapy (HRT). Estradiol works by restoring declining estrogen levels in the body, which helps alleviate distressing symptoms of menopause. However, it may affect the womb lining (endometrium) and increase the risk of endometrial cancer. Dydrogesterone is included to counteract the harmful effects of Estradiol on the womb lining and reduce the risk of cancer.
