• Serotonin Syndrome: A rare but potentially life-threatening condition presenting with agitation, hallucinations, rapid heart rate, fever, incoordination, and severe nausea/diarrhoea.

• Liver Problems: Unusual fatigue, abdominal pain, dark urine, or yellowing of the skin/eyes (jaundice).

• Severe Skin Reactions: Rash, blistering, or peeling skin.

Duloxetine is for oral use only and must be taken exactly as prescribed by your doctor.

• Swallow Whole: The capsule must be swallowed whole with a glass of water. Do not crush, chew, open, or dissolve the capsule, as this will damage the enteric coating and destroy the medication before it can be absorbed.

• Dosing Time: It is typically taken once daily, either in the morning or evening. Consistency is key, so take it at the same time each day.

• Food: The capsule can be taken with or without food.

• Anticoagulants/Antiplatelet Agents: Duloxetine can slightly impair platelet function, so combining it with warfarin, aspirin, or NSAIDs may increase the risk of bleeding.

This medicine should be used with extreme caution or is contraindicated in patients with the following health problems:

• Uncontrolled Narrow-Angle Glaucoma: Duloxetine can increase pupillary dilation, potentially worsening this condition.

• Severe Renal Impairment (Kidney Disease): Duloxetine is not recommended in patients with severe kidney disease, as it may accumulate in the body.

• End-Stage Liver Disease: Use is contraindicated in patients with significant liver impairment due to the drug's metabolism pathways.

• Severe Hypertension (Uncontrolled High Blood Pressure): Duloxetine can increase blood pressure. It should not be used until hypertension is controlled.

The starting dose for Duloxetine is typically 30mg once daily for one week, primarily for MDD and GAD. This lower dose allows the patient's body to adjust to the medication.

The dose is usually then increased to 60mg once daily for therapeutic efficacy. Doses for neuropathic pain and fibromyalgia may range from 60mg to a maximum of 120mg daily, based on individual response and tolerability, as determined by the prescribing physician.

An overdose of Duloxetine is a medical emergency.

Symptoms may include: Somnolence, serotonin syndrome, seizures, coma, tachycardia (fast heart rate), vomiting, and fainting.

If an overdose is suspected, seek emergency medical care immediately. Treatment involves supportive measures, monitoring vital signs, and managing serotonin syndrome symptoms.

If you miss a dose of Duloxetine :

Take the missed dose as soon as you remember.

If it is almost time for your next scheduled dose, skip the missed dose and continue with your regular dosing schedule.

Do not take two doses at the same time to make up for the missed one.
