Cefpodoxime Proxetil is a prodrug that converts into its active form Cefpodoxime, after absorption. Cefpodoxime binds to penicillin-binding proteins (PBPs) in bacterial cell walls, disrupting cell wall synthesis during bacterial growth and division. This leads to weakening of the bacterial cell wall, resulting in bacterial lysis and death (bactericidal action).

Cefpodoxime Proxetil is commonly prescribed for respiratory tract infections such as bronchitis, pneumonia, sinusitis, tonsillitis, and otitis media, as well as urinary tract infections and skin and soft tissue infections caused by organisms like Streptococcus, Haemophilus influenzae, E. coli, and Moraxella catarrhalis.

How it Works

Cefpodoxime Proxetil is a third-generation antibiotic which works by intefering into cell wall synthesis of bacteria, Cefpodoxime binds to penicillin-binding proteins (PBPs) involved in cross-linking of peptidoglycan, which are essential for forming the bacterial cell wall.

  • cephalosporins contains a beta-lactum ring by which it binds to PBPs (transpeptidases), preventing catalysis of cross-linking of peptidoglycan synthesis. peptidoglycan is a polymer formed in the bacterial cell wall, gives strength to bacteria, maintains shape and protects bacteria from lysis, cephalosporins works by inhibiting cross-linking to peptidoglycans.
  • Inhibition of cross-linking of peptidoglycans produces weak and unstable cell walls of bacteria.
  • The weakened wall can't withstand internal osmotic pressure, causing the cell to burst and die (cell lysis).

This mechanism effectively treats infections caused by Gram-positive bacteria such as Streptococcus and Gram-negative bacteria such as E. coli and Haemophilus influenzae. Since human cells do not have cell walls, Cefpodoxime selectively targets bacteria without harming normal body cells.
