How Amitriptyline and Ketamine work Amitriptyline and Ketamine is a combination of two medicines: Amitriptyline and Ketamine. Amitriptyline is a tricyclic antidepressant that increases the levels of chemical messengers (serotonin and noradrenaline) that inhibit the transmission of pain signals in the brain. Ketamine is a general anesthetic that works by blocking certain receptors (NMDA), thereby reducing the sensation of pain. Together, they effectively relieve neuropathic pain, which is pain resulting from damaged nerves.
