Vasograin Ec Tablet 20s is a four-component combination for the acute treatment of migraine and tension-type headaches. Ergotamine constricts widened (dilated) blood vessels inside your head (cranial blood vessels) to relieve throbbing pain, while caffeine enhances ergotamine's absorption, paracetamol provides analgesic relief, and prochlorperazine controls nausea and vomiting. Together, they tackle the pain, vascular, and gastrointestinal aspects of headache episodes in a single formulation.
This fixed-dose combination in Vasograin Ec Tablet 20s targets multiple aspects of headache pathophysiology all at once. Ergotamine acts as a non-selective agonist at serotonin, dopamine, and alpha-1 adrenergic receptors, narrowing dilated cranial blood vessels and providing relief. Caffeine amplifies ergotamine's vasoconstrictive effect and increases its oral absorption. Paracetamol inhibits prostaglandin synthesis, managing pain, while Prochlorperazine blocks dopamine D2 receptors in the brainstem to prevent nausea, vomiting, and sensory sensitivity during attacks.
This medicine is for short-term use and must be used when needed. It is most effective at the earliest onset of an attack. Overuse of Vasograin Ec Tablet 20s more than 2–3 days per week causes medication overuse headache (MOH). This medicine must not be taken during pregnancy, or by people suffering from cardiovascular disease, peripheral vascular disease, uncontrolled hypertension, or significant liver or kidney impairment.
Acute migraine with or without aura: To stop vascular headache at the earliest onset
For moderate-to-severe tension headache, particularly when accompanied by nausea
To manage headaches due to constriction of blood vessels in the cranium (head).
To manage gastrointestinal symptoms associated with migraine and to improve comfort and absorption
Vasograin Ec Tablet 20s is available as oral tablets. You can swallow it with a full glass of water, with or without food; however, a light meal may reduce stomach upset. Take at the first sign of a headache as it is most effective before peak intensity.
It must be taken as per your doctor’s direction. Do not exceed the prescribed maximum dose per attack or per week. Do not use more than 2 to 3 days per week to avoid MOH. Do not take Vasograin Ec Tablet 20s within 24 hours of any triptan. Do not combine it with other paracetamol-containing products.
Ergotamine acts on neurotransmitters (serotonin and dopamine) and alpha-1 receptors to relax widened blood vessels in the head and reduce migraine pain signals.
Caffeine increases ergotamine's effect of blood vessel constriction (vasoconstrictive effect) and also increases oral absorption by inhibiting first-pass metabolism. It also provides mild CNS stimulation that partially counteracts prochlorperazine's sedation.
When your body gets injured or infected, the cell membranes release a fatty acid called arachidonic acid. The COX enzyme converts that into prostaglandins. Prostaglandins are the chemicals that are responsible for pain, fever, swelling, and inflammation at the site. Paracetamol inhibits the cyclooxygenase (COX) pathway, reducing the production of prostaglandins, thereby decreasing pain perception.
Prochlorperazine blocks dopamine D2 receptors in the chemoreceptor trigger zone present in the brainstem (in medulla oblongata), preventing nausea and vomiting. It also provides sedation that reduces agitation and sensory sensitivity during attacks by blocking the Histamine H1-receptors.
Ergotamine can stimulate uterine contractions and hamper placental blood flow, causing fetal harm.
Ergotamine can pass into breast milk, causing adverse effects in infants. It may also suppress milk production.
Prochlorperazine causes significant drowsiness and impairs coordination. Avoid driving or operating heavy machinery.
People with an impaired liver may experience ergotism on continuous exposure. Paracetamol also carries a heightened hepatotoxicity risk in liver disease.
Ergotamine must not be used in severe renal impairment. Paracetamol dose adjustment may also be required.
Alcohol intake heightens the risk of paracetamol-induced liver toxicity and the likelihood of prochlorperazine's sedative and hypotensive effects.
Store at room temperature between 20-25°C in a dry place away from direct sunlight and moisture. Do not freeze. Keep out of reach of children. Do not use beyond the expiry date.
Triptans (like sumatriptan, rizatriptan, zolmitriptan) can cause additional vasospasm and increase the risk of cerebral or peripheral ischaemia
Medicines like erythromycin, ketoconazole & ritonavir block the metabolism of ergotamine, leading to fatal accumulation, vasospasm and ergotism.

Lowers ergotamine levels via CYP3A4 induction.
Increased risk of frequent seizure episodes.
Since this medicine is taken as needed at headache onset, a missed dose typically does not apply. If you have been recommended a scheduled regimen, take the missed dose as soon as you remember, unless the next dose is due within 2 hours. In such cases, it’s better to skip it than take double the dose.
